Gvardiol began playing football at the age of seven, when his father Tihomir, once an amateur player in his native Novigrad , took him to Trešnjevka. On 6 August 2023, Manchester City announced the signing of Joško Gvardiol until June 2028 from RB Leipzig for €90 million, making him the most expensive defender in football history. Gvardiol made his first appearance for the Citizens on 11 August 2023 in a 3–0 win over Burnley .
